Spectroscopy & Chromatography

This module covers instrumental structures of commonly used spectroscopic equipment, primarily Ultra Violet /Visible and Fourier transform infrared spectrometers will be described.

In addition, there is an introduction to the theory and instrumentation associated with Thin Layer Chromatography (TLC), Gas Chromatography (GC) and High Performance Liquid Chromatography (HPLC). 

Students will also learn how to carry out quantitative analyses associated with these techniques.

Spectroscopy

Single beam UV/Vis spectrometer. Light sources, monochromator, optics, detectors. Double beam instrument. Other means of background correction, Linear photodiode array detector, charge transfer devices. Interferometry. Infrared sources, detectors, optics, relation between interferogram and spectrum. Deviations from Beers law. Zeroing the instrument, use of blanks, data interpretation, working with concentration units, applying a dilution factor

Chromatography

Introduction to Liquid-liquid extraction. Distribution coefficient. Classification of chromatography. Theory of planar chromatography, thin layer chromatography, paper chromatography, Rf values. Comparison of planar chromatography and column chromatography. Description of GC instrumentation: modes of injection, stationary phases, columns- packed and capillary, detectors High Performance Liquid chromatography, normal phase and reverse phase. Chromatogram interpretation. Qualitative analysis using GC and HPLC. Choice of sustainable mobile phase.There will be a range of suitable practicals available, which support the theory.

The module is delivered through a series of lectures (20 hours), tutorials (4 hours), laboratory practical classes (15 hours) and self-study (61 hours).
